T-cell dependent immunoselection

چکیده
In a recent issue of Nature, two groups describe the process of cancer immunoediting whereby an individual is protected from cancer though the elimination and immunogenic modification of cancer cells. Matsushita and colleagues use whole-exon sequencing of chemically induced mouse sarcomas to obtain evidence for T cell-mediated immunoselection. The authors characterized mutations in immunodeficient mice and thereby elegantly identified a tumor rejection antigen. Further it was demonstrated that T cell-dependent immunoselection is a crucial mechanism underlying the outgrowth of tumor cells. This exciting study was published alongside a study from DuPage et al. demonstrating that immunosurveillance and immunoediting can occur in an oncogene-driven endogenous tumor model provided that the tumors carry strong neo-antigens that are not present in the host. DuPage and colleagues showed that loss of tumor antigen expression or presentation on Class I MHC molecules was necessary and T-cell dependent immunoselection

Efficient chromosomal mapping of a methylcholanthrene-induced tumor antigen by CTL immunoselection.
It has been difficult to genetically map the genes encoding tumor Ags because they arise as a consequence of somatic mutational events. CTL-mediated immunoselection can impose potent immunoselective pressure against tumor cells, resulting in the survival of rare tumor Ag-loss variants. We subjected a heterozygous 3-methylcholanthrene-induced murine sarcoma cell line to CTL immunoselection, sele...

Immunoselection via complement-dependent lysis of human-mouse somatic cell hybrids containing chromosome 7, with antisera reactive to cell surface antigen(s) coded for by chromosome 7, has resulted in growth of somatic cell hybrids containing rearranged human chromosome 7s. Investigation of these hybrids has localized the gene(s) coding for the relevant cell surface antigen(s) to the short arm ...
متن کاملLoss of a unique tumor antigen by cytotoxic T lymphocyte immunoselection from a 3-methylcholanthrene-induced mouse sarcoma reveals secondary unique and shared antigens
Most chemically induced tumors of mice express unique antigens that can be recognized by cytotoxic T lymphocytes (CTL) and thereby mediate tumor rejection. The number of different antigens expressed by a single tumor and their interplay during immunization and rejection are largely unexplored. We used CTL clones specific to individual tumor antigens to examine the number and distribution of CTL...
متن کاملGeneration of T cells specific for the wild-type sequence p53(264-272) peptide in cancer patients: implications for immunoselection of epitope loss variants.
Alterations in the p53 gene occur frequently and can lead to accumulation of p53 protein in squamous cell carcinomas of the head and neck (SCCHN). Since accumulation of p53 is associated with enhanced presentation of wild-type sequence (wt) p53 peptides to immune cells, the development of pan vaccines against SCCHN has focused on wt p53 epitopes. We used the HLA-A2.1-restricted wt p53(264-272) ...
متن کاملKinetics of in vivo elimination of suicide gene-expressing T cells affects engraftment, graft-versus-host disease, and graft-versus-leukemia after allogeneic bone marrow transplantation.
Suicide gene therapy is one approach being evaluated for the control of graft-vs-host disease (GVHD) after allogeneic bone marrow transplantation (BMT). We recently constructed a novel chimeric suicide gene in which the entire coding region of HSV thymidine kinase (HSV-tk) was fused in-frame to the extracellular and transmembrane domains of human CD34 (DeltaCD34-tk). DeltaCD34-tk is an attracti...
